To check this, we additionally optimized the structures of the L2, L6 ligands by means of more accurate ab initio unrestricted HF ; method with the 6-311G * basis. The new structures appeared very similar to those obtained by DFT with B3LYP potential. Therefore, we may expect that the non-planar structures of the chelate rings are not artifacts, and the planarity of the sulphur-containing chelate ring in the crystal results from interactions between the chelate molecules in the solid state. Supplementary calculations on this hypothetical "packing" effect, carried out for systems consisting of two, three and four [Tc CO ; 3L6Cl] molecules, indicate that with increasing number of closely packed chelate molecules both the deformation of the chelate ring and the length of the Tc-S bond decrease. Table 2 presents Mulliken charge distribution on the metal and the O and S donor ligand atoms in the [Tc CO ; 3LCl] chelates studied. Charge transfer from the ligands to the central metal ion upon chelate formation, decreases the positive charge on Tc and increases the negative charge on both donor atoms in the coordinated N, O-ligands, just as reported in [4]. The charges on the N1 donor atoms in the chelates with the five-membered rings are nearly constant, qN1 -0.22 to -0.24 e, as well as the charges on the Cl atoms in the chelates, qCl -0.32 to -0.34 e, except -0.41 e for L7. In the case of the N, S-ligand chelates a moderate increase in the negative charge on the N1 atoms is also observed, however, the decrease in the positive charge on the metal is large, at the expense of large changes in qS. Characteristics of Society Members with a Systems Orientation Analysis of orientation trends during the past 25 years suggests that systems models are increasingly adopted by pediatric psychologists. Over half n 27; 68% ; of the respondents endorsing a primary systems orientation graduated after 1980, with less than one third of the respondents with a systems orientation n 13; 32% ; graduating between 1965 and 1980. Assessing graduation dates in 5-year increments over the last 25 years, less than 5% n 3 ; of the total 19701974 graduates espoused a systems orientation, whereas 13.3% n 16 ; of the total number of 1986-1990 graduates endorsed a systems approach. Of the 40 pediatric psychologists who reported having a systems orientation, 40% n 16 ; were employed in either medical school or traditional academic settings. Twenty-eight percent n 11 ; of the respondents were private practitioners; 5% n 2 ; endorsed more than one employment setting; and one was employed in a community mental health center. With respect to research, over half of the systems-oriented pediatric psychologists n 26; 65% ; stated that they currently were involved in empirical endeavors. Of these respondents, approximately 45% n 12 ; reported that their research efforts involved the direct implementation of systems models. This stands in contrast to respondents endorsing a cognitive-behavioral orientation, who stated that family systems models were used in only 8.4% of their research. Only6.1% 30 ; of the entire sample believed that systems theory principles could be studied empirically and pegvisomant.

DISCUSSION The camptothecin analog irinotecan has shown substantial antitumor activity against syngeneic and heterotransplant models, notably xenograft tumors derived from childhood cancers 57 ; . Early clinical results also indicate a high level of activity for irinotecan with marked antitumor activity in phase I trials in several histologies 12 ; as well as in advanced-stage rhabdomyosarcoma at diagnosis 41 ; . Because it has been reported that the epidermal growth factor receptor ERBB1 ; is expressed in several pediatric tumor types, including neuroblastoma, rhabdomyosarcoma, and glioblastoma, we have examined the antitumor activity of the ERBB1 inhibitor, gefitinib, alone or in combination with irinotecan. As a single agent administered either one or two times daily by oral gavage, gefitinib showed no substantial antitumor activity against any of the 10 tumor lines examined. In contrast, gefitinib significantly potentiated the antitumor activity of irinotecan in four tumor lines SJ-GBM2, NB-1691, NB-1382, Rh28.

Ediatric vasculitis is a diagnostic challenge. However, a new international classification for the vasculitides observed in children promises a more specific and realistic guideline criteria, bringing it in line with current clinical practice. "The new classification is detailed and represents the amalgamation of what has been an understanding of how vasculitis should be classified in children for quite a long time, " said Professor Taunton Southwood, a pediatric rheumatologist at the University of Birmingham, UK. The new classification was presented in April and endorsed by the European League Against Rheumatism and the Paediatric Rheumatology European Society. This has led to the craze of Botox parties in which several people gather for Botox treatments in an effort to eliminate wasting the product. Botox also is approved by the FDA to treat several medical conditions its "therapeutic" applications ; . These include cervical dystonia involuntary movement and prolonged muscle contraction of the neck ; , blepharospasm involuntary forcible closure of the eyelids ; , strabismus crossed eyes ; , hyperhidrosis excessive sweating ; , and dynamic muscle contracture in pediatric cerebral palsy patients an abnormality of motor function usually acquired at a young age ; . Botox is sometimes used for other medical conditions without specific FDA approval so-called "offlabel" applications which the company cannot advertise ; . Botox designated for wrinkle smoothing is marketed as BOTOX Cosmetic, whereas the product destined for medical applications is simply called Botox or Botox Therapeutic. Are There Promising Alternatives to the LD50 Test for Botox? Replacement Alternatives: Several approaches to assessing the potency of Botulinum toxin are potential candidates for replacing the LD50 Test. While the LD50 Test dates back to the 1920s, the potential replacements all take advantage of more modern technology and insights into the mechanism by which Botulinum toxin exerts its effects on the body. Perhaps the most promising replacement is the endopeptidase assay. Modern research has shown that Botulinum toxin type A the key ingredient in Botox ; exerts its paralyzing effect by disrupting a molecule --SNAP25-- critical in transmitting nerve signals to muscles. The endopeptidase assay evaluates the ability of toxin samples to disrupt this molecule, using the latest tools of molecular biology and immunology. The assay's developers suggest that it could replace the mouse assay in Botulinum toxin testing. Reduction Alternatives: Some studies have looked at ways to reduce the number of animals used in the LD50 Test for Botulinum potency conventionally 100 animals ; . Pearce and colleagues assessed Botulinum toxin potency using various numbers of animals, including 100, 50, and 25, and found excellent results for the 50 and 25 animal tests. They concluded that the 25-animal assay, using 5 mice per dose, may be adequate for most laboratory experimentation when only a single determination of the LD50 is made. Allergan itself, the manufacturer of Botox, may have found similar results, as the company claims to have reduced animal numbers per test by 50% and is preparing to implement another 50% reduction, once approved by the U.S. FDA, though the company has not revealed the actual numbers of animals involved. Refinement Alternative: Various animal-based tests have been proposed as substitutes for the LD50 Test in assessing Botulinum toxin potency, all of which cause less animal suffering and are more technologically sophisticated than the LD50 Test. Instead of injecting high doses of toxin to cause wholesale paralysis and death, the refinement alternatives involve administering low doses to assess localized or regional effects. Perhaps the most promising is the mouse hind limb assay, in which the toxin is injected into the calf muscle. Complete paralysis of the injected limb results in two days.
